From Antibodies.com, this competitive enzyme-linked immunosorbent assay (cELISA) is designed for the in vitro quantitative determination of cortisol in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated microplate arranged as 12 x 8 well strips, in which cortisol present in samples or standards competes with immobilised cortisol antigen for binding sites on a biotinylated anti-cortisol antibody. Following incubation and washing, an HRP-avidin conjugate is added and a TMB substrate is used to visualise the reaction; the resulting yellow colour is inversely proportional to the amount of cortisol in each sample or standard and is read at 450 nm on a colorimetric microplate reader.
The kit covers a working range of 1.56–100 ng/ml with a sensitivity of 0.62 ng/ml and a total assay time of 2 hours 30 minutes. Kit components are stored at +4°C or −20°C as instructed. Reported recoveries (n=5) are 88–102% (average 95%) for serum, 90–100% (average 95%) for EDTA plasma and 90–102% (average 96%) for heparin plasma. Dilution linearity has been characterised at 1:2, 1:4, 1:8 and 1:16 dilutions for each of these sample matrices.
