General Carboxymethyl Lysine ELISA Kit

From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is intended for the in vitro quantitative determination of Carboxymethyl Lysine in serum, plasma, tissue homogenates or other biological fluids. The kit uses a 96-well microtiter plate pre-coated with Carboxymethyl Lysine antigen and the competitive enzyme immunoassay technique.

During incubation, Carboxymethyl Lysine present in samples or standards competes with the immobilised antigen for binding sites on a Biotinylated Anti-Carboxymethyl Lysine Antibody. After washing, an HRP-Avidin conjugate is added, followed by TMB substrate solution. The resulting blue-coloured product changes to yellow upon addition of acidic stop solution, with the intensity of the yellow colour inversely proportional to the amount of Carboxymethyl Lysine in each well. Concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a range of 78.1-5000 ng/ml and a sensitivity of <33 ng/ml, with a total assay time of approximately 2 h 30 min. Reported recovery values are 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma and 80%-89% (average 84%) for heparin plasma. Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past the stated expiration date.
