General Advanced Glycation End Product ELISA Kit

From Antibodies.com, this competitive enzyme-linked immunosorbent assay (cELISA) is designed for the in vitro quantitative determination of Advanced Glycation End Products (AGE) in serum, plasma, tissue homogenates or other biological fluids. The assay employs a 96-well microtiter plate pre-coated with AGE antigen, in which AGE present in samples or standards competes with the immobilised antigen for binding sites on a Biotinylated Anti-AGE Antibody. An HRP-Avidin conjugate is then added, and TMB substrate is used to visualise the reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour is inversely proportional to the amount of AGE present, and the concentration is calculated by reading the absorbance at 450nm on a colorimetric microplate reader.

Reported performance characteristics include a product range of 125-8000 ng/ml and a sensitivity of <44 ng/ml, with a total assay time of 2h 30m. The kit has been validated using serum, EDTA plasma and heparin plasma samples. Recovery in serum (n=5) was 80%-102% (average 91%); in EDTA plasma (n=5) 81%-100% (average 90%); and in heparin plasma (n=5) 80%-89% (average 84%). Linearity has been assessed across dilution series from 1:2 to 1:16. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
