From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is intended for the in vitro quantitative determination of horse Matrix Metalloproteinase 1 (MMP1) in serum, plasma, tissue homogenates or other biological fluids. An antibody specific for MMP1 is pre-coated onto a microplate. Standards and samples are added to the wells and any MMP1 present is captured by the immobilized antibody. After incubation and washing, a Biotinylated Anti-MMP1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of the acidic stop solution. The intensity of yellow is proportional to the amount of MMP1 captured in each well, and the concentration is calculated from the O.D. absorbance at 450 nm in a microplate reader by reference to a standard curve.

The assay reports a sensitivity of 0.040 ng/ml and a working range of 0.156-10 ng/ml, with a total assay time of 4h 30m. Sample recovery is reported for serum at 80%-102% with an average of 91% (n=5), for EDTA plasma at 81%-100% with an average of 90% (n=5), and for heparin plasma at 80%-89% with an average of 84% (n=5). Linearity data are provided across serial dilutions from 1:2 to 1:16 in serum, EDTA plasma and heparin plasma. The platform is a pre-coated microplate configured as 12 x 8 well strips and is read on a colorimetric microplate reader.

Kit components include a pre-coated 96 well microplate, lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date. The assay is reactive with equine (horse) samples.
