Equine Interleukin 8 ELISA Kit

From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative measurement of equine interleukin-8 (IL-8) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The procedure uses a pre-coated microplate with an immobilised anti-IL-8 capture antibody, followed by a biotinylated anti-IL-8 detection antibody, HRP-avidin and TMB substrate; absorbance is read at 450 nm. Detection is colorimetric and the total assay time is 4 h 30 min.

Performance figures quoted by the manufacturer include a working range of 31.2-2000 pg/ml and a sensitivity of <12.4 pg/ml. Spiked recovery in neat samples averaged 91% in serum (n=5, range 80-102%), 90% in EDTA plasma (n=5, range 81-100%) and 84% in heparin plasma (n=5, range 80-89%). Dilution linearity (n=5) at 1:2, 1:4, 1:8 and 1:16 dilutions gave recoveries of 87-91%, 87-107%, 74-101% and 92-97% in serum; 90-105%, 84-101%, 90-101% and 79-108% in EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% in heparin plasma.

Kit components comprise a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage conditions split between +4°C and -20°C depending on the component. Third-party documentation lists the detection antibody host as rabbit (IgG), supplied in PBS with 0.02% sodium azide and 50% glycerol at pH 7.3, and recommends storage at 4°C while avoiding freeze/thaw cycles.
