Equine Interleukin 10 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com designed for the in vitro quantitative determination of horse Interleukin 10 (IL-10) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay uses a pre-coated 96-well microplate in a 12 x 8 well strip format. Standards and samples are added to wells where IL-10 binds to the immobilized capture antibody, followed by incubation with a Biotinylated Anti-IL-10 Antibody and then an HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm on a colorimetric microplate reader and IL-10 concentration is calculated from the standard curve. Total assay time is 4h 30m, with a measuring range of 15.6-1000 pg/ml and a sensitivity of <5.4 pg/ml. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

Spike-and-recovery performance: serum range 83-100% (average 91%, n=5), EDTA plasma 90-103% (average 96%, n=5) and heparin plasma 89-102% (average 95%, n=5). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 has been demonstrated in serum (n=5: 83-110%, 79-100%, 90-98%, 82-108%), EDTA plasma (n=5: 90-98%, 90-105%, 73-109%, 87-91%) and heparin plasma (n=5: 88-103%, 77-105%, 95-98%, 87-110%).
