Chicken Interleukin 2 ELISA Kit

Antibodies.com provides a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of chicken IL-2 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips and produces a colorimetric readout measured at 450 nm.

The kit employs the sandwich enzyme immunoassay technique. Standards and test samples are added to wells pre-coated with an antibody specific for IL-2, allowing IL-2 present in each sample to bind to the immobilised capture antibody. Following washing, a biotinylated anti-IL-2 antibody is added, which binds the captured IL-2, followed by incubation with HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of IL-2 captured per well, and the concentration of IL-2 is calculated by reference to a standard curve.

The quantitative measurement range is 15.6-1000 pg/ml with a sensitivity of <6.5 pg/ml. Total assay time is 4h 30m. Reported neat-sample recovery ranges are 80-102% for serum (n=5, average 91%), 81-100% for EDTA plasma (n=5, average 90%) and 80-89% for heparin plasma (n=5, average 84%). Validated dilution factors include 1:2, 1:4, 1:8 and 1:16. Kit components are stored at +4°C or -20°C as instructed in the protocol, and the kit should not be used past its expiration date.
