Manufactured by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of chicken Brain Derived Neurotrophic Factor (BDNF). The assay is reactive with chicken samples and accepts serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. Detection is colourimetric, read on a standard microplate reader at 450 nm, and the procedure uses a pre-coated 96-well microplate configured as 12 x 8 well strips.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for BDNF is pre-coated onto the microplate; standards and test samples are added and BDNF present in each sample is bound by the immobilised antibody. After washing, a Biotinylated Anti-BDNF Antibody binds the captured analyte, followed by HRP-Avidin conjugate and TMB substrate. The resulting blue product turns yellow upon addition of acidic stop solution, with absorbance at 450 nm proportional to the amount of BDNF captured in each well and quantified by reference to the standard curve.

Quantitative performance characteristics quoted by the manufacturer include a product range of 0.156-10 ng/ml, a sensitivity of <0.065 ng/ml, and a total assay time of 4h 30m. Reported recovery in spiked samples averages 91% in serum (range 80%-102%, n=5), 90% in EDTA plasma (range 81%-100%, n=5), and 84% in heparin plasma (range 80%-89%, n=5). Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
