Canine Thrombopoietin ELISA Kit

From Antibodies.com, this sandwich enzyme-linked immunosorbent assay (sELISA) enables the in vitro quantitative determination of canine Thrombopoietin in serum, plasma, or other biological fluids. An antibody specific for Thrombopoietin is pre-coated onto the microplate, and standards and test samples are added to the wells where Thrombopoietin is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-Thrombopoietin Antibody binds the captured analyte, after which an HRP-Avidin conjugate is added. TMB substrate then visualises the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the Thrombopoietin concentration is calculated by reference to a standard curve.

The assay has a sensitivity of <6.9 pg/ml and a quantitative range of 15.6-1000 pg/ml, with a total assay time of 4h 30m. Recovery testing gave the following results: serum (n=5) 80% - 102% (average 91%); EDTA plasma (n=5) 81% - 99% (average 90%); heparin plasma (n=5) 80% - 89% (average 84%). Dilutional linearity was assessed at 1:2, 1:4, 1:8, and 1:16 dilutions across all three sample types.

The kit is supplied with a pre-coated microplate, lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
