Description

From Antibodies.com, this sandwich-format Enzyme-Linked Immunosorbent Assay is designed for the in vitro quantitative determination of canine Thrombomodulin in plasma. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for Thrombomodulin immobilised on the plate. Standards and test samples are added to the wells, and captured Thrombomodulin is detected using a Biotinylated Anti-Thrombomodulin Antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate solution, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of Thrombomodulin captured in each well, and the concentration is calculated by reading absorbance at 450 nm on a colorimetric microplate reader and referring to the standard curve.

The quoted assay range is 15.6–1000 pg/ml with a sensitivity of less than 5.8 pg/ml. Total assay time is 4 hours 30 minutes. Recovery has been characterised in serum (average 92%, range 82%–102%, n=5), EDTA plasma (average 95%, range 90%–100%, n=5), and heparin plasma (average 94%, range 88%–100%, n=5). Linearity on dilution has been reported at 1:2, 1:4, 1:8, and 1:16 across the same three sample types, with corresponding recovery ranges reported for each dilution.

Kit components should be stored at +4°C or −20°C as instructed and the kit should not be used past its expiration date. The assay is intended for use with plasma samples.
