Description

This competitive Enzyme-Linked Immunosorbent Assay (cELISA) from Antibodies.com is designed for the in vitro quantitative determination of canine Parathyroid Hormone in serum, plasma or other biological fluids. The kit uses a competitive enzyme immunoassay technique with a 96-well microtiter plate pre-coated with Parathyroid Hormone antigen. During incubation, Parathyroid Hormone present in samples or standards competes with the fixed amount of immobilized antigen for binding sites on a Biotinylated Anti-Parathyroid Hormone Antibody. After washing to remove unbound antibody, an HRP-Avidin conjugate is added. Following further incubation and washing, TMB substrate solution produces a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is inversely proportional to the Parathyroid Hormone concentration, which is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to a standard curve.

The assay has a stated sensitivity of <3.52 pg/ml and a measuring range of 12.5–800 pg/ml. Detection is colorimetric, performed on a microplate reader, and the total assay time is approximately 2 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.

Recovery has been characterised across five samples per matrix: serum (average 91%, range 80–102%), EDTA plasma (average 90%, range 81–99%) and heparin plasma (average 84%, range 80–89%).
