From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of canine Osteopontin in serum, plasma, tissue homogenates or other biological fluids. The assay employs pre-coated microplate wells (12 x 8 well strips) with an antibody specific for Osteopontin. Standards and samples are added to the wells and captured Osteopontin is detected using a Biotinylated Anti-Osteopontin Antibody followed by HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of stop solution. The yellow colour density, proportional to the Osteopontin concentration, is read as optical density at 450 nm using a colorimetric microplate reader.
The assay has a reported sensitivity of <0.61 ng/ml and a working range of 1.56-100 ng/ml, with a total assay time of 4h 30m. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
Reported spike-and-recovery averages in the validation data are 91% in serum, 90% in EDTA plasma, and 84% in heparin plasma, with dilution linearity reported across 1:2 to 1:16 dilutions for each sample type.
