This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of canine inducible nitric oxide synthase (iNOS) in serum, plasma, tissue homogenates and other biological fluids. An antibody specific for iNOS is pre-coated onto the microplate; standards and samples are added, washed and detected with a biotinylated anti-iNOS antibody, HRP-avidin conjugate and TMB substrate. The enzymatic reaction is visualised by a blue-to-yellow colour change and read at 450 nm on a colorimetric microplate reader, with iNOS concentration calculated against the standard curve.

According to the manufacturer, the kit has a sensitivity of <0.126 ng/ml and an assay range of 0.312–20 ng/ml. Total assay time is 4h 30m on a pre-coated microplate arranged as 12 x 8 well strips, and components are stored at +4°C or -20°C as instructed.

Manufacturer-reported recovery averages across undiluted samples are 91% for serum (n=5, range 80–102%), 90% for EDTA plasma (n=5, range 81–100%) and 84% for heparin plasma (n=5, range 80–89%). Dilution linearity data show serum recovery of 87–91% (1:2), 87–107% (1:4), 74–101% (1:8) and 92–97% (1:16), EDTA plasma of 90–105%, 84–101%, 90–101% and 79–108%, and heparin plasma of 84–95%, 92–105%, 82–105% and 89–91%.
