The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate. An antibody specific for Lactoferrin captures the analyte in standards and test samples, with detection carried out using a Biotinylated Anti-Lactoferrin Antibody and an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a colour change that is read at 450 nm in a microplate reader, and the Lactoferrin concentration is calculated by reference to a standard curve.

The assay covers a product range of 31.2–2000 pg/ml with a sensitivity of <9.27 pg/ml, and the total assay time is 4 h 30 min. Reported spike-and-recovery averages (n=5) are 91% for serum (range 80–102%), 90% for EDTA plasma (81–99%) and 84% for heparin plasma (80–89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions yielded recoveries of 87–107% for serum, 79–108% for EDTA plasma and 82–105% for heparin plasma.
