Canine Interleukin 4 ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of canine Interleukin 4 (IL-4) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. The kit employs a 96-well microtiter plate pre-coated with an antibody specific for IL-4. Standards and samples are added to the wells, where IL-4 is captured by the immobilised antibody. Following incubation and washing, a biotinylated anti-IL-4 detection antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader.

The assay has a sensitivity of 6.5 pg/ml and a working range of 15.6-1000 pg/ml, with a total assay time of 4 hours 30 minutes. Spike-and-recovery values are given for serum (80-102%, average 91%), EDTA plasma (81-99%, average 90%), and heparin plasma (80-89%, average 84%), with linearity demonstrated across serial dilutions from 1:2 to 1:16. Kit components include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, to be stored at +4°C or -20°C as instructed.
