Description

Designed for the in vitro quantitative determination of canine IP10 (Interferon Gamma Induced Protein 10kDa), this sandwich ELISA from Antibodies.com is intended for serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips; standards and samples are added to the wells and the IP10 present is captured by the immobilised antibody. Following incubation and washing, a biotinylated anti-IP10 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow on addition of acidic stop solution, and the absorbance is read at 450 nm in a microplate reader.

The standard curve covers a range of 15.6-1000 pg/ml, with a sensitivity of <6.8 pg/ml, and the complete assay can be performed in 4h 30m. Reported recovery in neat serum averages 91% (range 80%-102%, n=5), in EDTA plasma averages 90% (81%-100%, n=5), and in heparin plasma averages 84% (80%-89%, n=5). Dilution linearity has been assessed at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma samples (n=5 each). Kit components should be stored at +4°C or -20°C as instructed and the product should not be used past its expiration date.
