Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of canine Intercellular Adhesion Molecule 1 (ICAM1) in serum, plasma, tissue homogenates or other biological fluids. The kit employs a pre-coated 96-well microplate configured as 12 x 8 well strips, with an antibody specific for ICAM1 immobilised on the wells.

Standards and test samples are added to the wells, where ICAM1 is captured by the immobilised antibody. After washing, a Biotinylated Anti-ICAM1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of ICAM1 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve. The assay has a reported sensitivity of <0.064 ng/ml and a measuring range of 0.156–10 ng/ml, with a total assay time of 4h 30m.

The kit is suitable for serum, plasma and tissue homogenates. Reported recovery performance includes serum (average 91%, range 80%–102%), EDTA plasma (average 90%, range 81%–100%) and heparin plasma (average 84%, range 80%–89%). Kit components should be stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
