From Antibodies.com, this Sandwich ELISA kit is designed for the in vitro quantitative determination of canine Insulin Like Growth Factor 1 (IGF1) in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for IGF1 immobilised on a 96-well microplate. Standards and samples are added to the wells; after washing, a Biotinylated Anti-IGF1 Antibody is applied, followed by an HRP-Avidin conjugate. TMB substrate is then added, producing a blue colour that turns yellow upon addition of the stop solution. Absorbance is read at 450 nm in a microplate reader and IGF1 concentration is calculated against the standard curve.

The kit has an assay range of 2.5–160 ng/ml and a sensitivity of <0.9 ng/ml, with a total assay time of 4 h 30 min on a pre-coated microplate platform (12 x 8 well strips). Detection is colourimetric. Reported recovery: serum (n=5) range 80%–102%, average 91%; EDTA plasma (n=5) range 81%–100%, average 90%; heparin plasma (n=5) range 80%–89%, average 84%. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is given for serum (87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (84–95%, 92–105%, 82–105%, 89–91%), all n=5. Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or −20°C as instructed and the kit must not be used past its expiration date.
