Canine Endothelin 1 ELISA Kit

A competitive Enzyme-Linked Immunosorbent Assay (cELISA) from Antibodies.com designed for the in vitro quantitative determination of canine Endothelin 1 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay uses the competitive enzyme immunoassay technique on a pre-coated microplate. Endothelin 1 in samples or standards competes with the immobilized antigen for binding sites on a biotinylated anti-Endothelin 1 antibody; the more analyte present, the less antibody binds to the plate. After washing, an HRP-Avidin conjugate is added and detection is achieved with a TMB substrate, producing a blue product that turns yellow on addition of an acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, with the density of yellow inversely proportional to the amount of Endothelin 1 in each sample or standard. Reported sensitivity is <2.33 pg/ml and the quantitative range is 7.81–500 pg/ml; total assay time is approximately 2 hours 30 minutes.

Recovery has been characterised in undiluted samples (serum 80–102%, average 91%; EDTA plasma 81–100%, average 90%; heparin plasma 80–89%, average 84%) and across serial dilutions from 1:2 to 1:16 in serum, EDTA plasma and heparin plasma. Kit components should be stored at +4°C or −20°C as instructed, and the product should not be used past its expiration date.
