From Antibodies.com, this Sandwich ELISA kit is designed for the in vitro quantitative determination of canine COL1A2 in serum, plasma or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, with an antibody specific for COL1A2 pre-coated onto a microplate. Standards and test samples are added to the wells, and COL1A2 present in each sample is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-COL1A2 Antibody binds the captured analyte, and an HRP-Avidin conjugate is added. TMB substrate visualises the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of COL1A2 captured in each well, and concentration is calculated from O.D. absorbance at 450 nm in a microplate reader, referenced to a standard curve.

The assay offers a sensitivity of <0.126 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of 4h 30m. Recovery performance was characterised with n=5 for each sample type: serum ranged 83%–100% with an average of 91%; EDTA plasma ranged 90%–103% with an average of 96%; heparin plasma ranged 89%–102% with an average of 95%. Dilution linearity was tested at 1:2, 1:4, 1:8, and 1:16 across serum, EDTA plasma, and heparin plasma. Kit components are stored at +4°C or -20°C as instructed, and the product must not be used past its expiration date.

Detection is colourimetric on a pre-coated microplate platform, and the assay is validated for the quantitative measurement of Collagen Type I Alpha 2 in canine samples.
