Antibodies.com offers a sandwich enzyme-linked immunosorbent assay (sELISA) kit for the in vitro quantitative determination of canine Coagulation Factor VIII in plasma or other biological fluids. The assay uses a pre-coated microplate with antibody specific for Factor VIII, a biotinylated anti-Factor VIII detection antibody, an HRP-avidin conjugate, and TMB substrate, with absorbance read at 450 nm on a colorimetric microplate reader.

The standard curve range is 3.12–200 ng/ml with a sensitivity of <1.15 ng/ml. Total assay time is 4 hours 30 minutes. In spike-and-recovery testing (n=5), average values were 92% for serum, 95% for EDTA plasma, and 94% for heparin plasma, with corresponding individual ranges of 82–102%, 90–100%, and 88–100% respectively. Dilution linearity was evaluated at 1:2, 1:4, 1:8, and 1:16 in each matrix (n=5), producing the following recovery windows: serum 86–110%, 77–102%, 90–96%, 84–108%; EDTA plasma 88–100%, 90–103%, 76–110%, 86–93%; heparin plasma 89–102%, 75–106%, 94–97%, 88–110%.

Components supplied include the pre-coated microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, with storage temperatures specified at +4°C and -20°C. The kit should not be used past its expiration date.
