Description

A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of canine TRAP/CD40L (Cluster of Differentiation 40 Ligand) in serum, plasma, tissue homogenates or other biological fluids. The kit is intended for quantitative measurement of the target in canine samples.

The assay uses a pre-coated microplate in a 12 x 8 well strip format with a colourimetric detection method read at 450 nm. Standards and samples are incubated with an immobilised anti-TRAP/CD40L capture antibody, followed by a biotinylated anti-TRAP/CD40L antibody and an HRP-avidin conjugate. TMB substrate is then added; the enzymatic reaction produces a blue product that turns yellow upon addition of stop solution, and the intensity of yellow is proportional to the amount of TRAP/CD40L captured in each well. Total assay time is 4 hours 30 minutes. The assay range is 0.156–10 ng/ml with a reported sensitivity of <0.032 ng/ml.

Spike-and-recovery performance was reported as: serum (n=5) range 83%–100% with an average of 91%; EDTA plasma (n=5) range 91%–98% with an average of 94%; heparin plasma (n=5) range 90%–99% with an average of 94%. Dilution linearity data are provided across 1:2, 1:4, 1:8, and 1:16 dilutions for serum, EDTA plasma, and heparin plasma. Kit components should be stored at +4°C or −20°C as instructed, and the product should not be used past its expiration date.
