Description
This sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of canine Calcitonin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated microplate platform with colorimetric detection, read on a microplate reader at 450 nm. In the sandwich immunoassay format, an antibody specific for Calcitonin is immobilised on the plate; standards and samples are added and Calcitonin is captured, then the wells are washed and incubated with a biotinylated anti-Calcitonin antibody. HRP-Avidin conjugate is added next, followed by TMB substrate, which produces a blue product that turns yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of Calcitonin captured in that well, and the concentration is calculated against a standard curve.
According to the manufacturer's documentation, the stated range is 15.6–1000 pg/ml and sensitivity is <7.7 pg/ml. Typical assay time is 4 hours 30 minutes. Recovery data supplied with the kit: serum (n=5, range 82–102%, average 92%), EDTA plasma (n=5, 90–100%, 95%) and heparin plasma (n=5, 88–100%, 94%). Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
