Description

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of bovine TREM1 (Triggering Receptor Expressed On Myeloid Cells 1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay has a working range of 15.6-1000 pg/ml and a sensitivity of <5.4 pg/ml, with absorbance read on a colorimetric microplate reader at 450 nm. Total assay time is 4 hours 30 minutes.

Validated spike-and-recovery values include 80-102% in serum (average 91%), 81-100% in EDTA plasma (average 90%), and 80-89% in heparin plasma (average 84%). Dilution linearity at 1:2, 1:4, 1:8, and 1:16 ranged from 87-91%, 87-107%, 74-101%, and 92-97% in serum; 90-105%, 84-101%, 90-101%, and 79-108% in EDTA plasma; and 84-95%, 92-105%, 82-105%, and 89-91% in heparin plasma.

The kit uses a pre-coated 96-well microplate. Standards and samples are added to the wells, where TREM1 is captured by the immobilized antibody. After incubation and washing, a biotinylated anti-TREM1 antibody binds the captured analyte, followed by HRP-avidin conjugate and TMB substrate. The enzymatic reaction yields a blue product that turns yellow upon addition of acidic stop solution; absorbance at 450 nm is proportional to TREM1 concentration and is calculated against a standard curve. Components include the pre-coated microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, stored at +4°C or -20°C as instructed.
