Bovine Tissue Inhibitors Of Metalloproteinase 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, the kit is designed for the in vitro quantitative determination of bovine Tissue Inhibitors of Metalloproteinase 2 (TIMP2) in serum, plasma, or other biological fluids. The assay uses a pre-coated 96-well microtiter plate on which an antibody specific for TIMP2 is immobilised. Standards and samples are added to the wells, where any TIMP2 present is captured by the immobilised antibody. After washing, a Biotinylated Anti-TIMP2 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow on addition of stop solution; the intensity of this yellow colour is proportional to the amount of TIMP2 captured in each well. Concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to a standard curve.

The quantitative assay range is 62.5-4000 pg/ml with a sensitivity of 22.3 pg/ml. Reported recovery averages 91% in serum (range 80%-102%, n=5), 90% in EDTA plasma (range 81%-100%, n=5) and 84% in heparin plasma (range 80%-89%, n=5). Total assay time is approximately 4 hours 30 minutes, and detection is colorimetric on a standard microplate reader. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
