From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of bovine Tissue Inhibitors Of Metalloproteinase 1 (TIMP1) in serum, plasma or other biological fluids. The assay employs a sandwich enzyme immunoassay technique in which a TIMP1-specific antibody is pre-coated onto a 96-well microplate. Standards and samples are added to the wells, and captured TIMP1 is detected using a Biotinylated Anti-TIMP1 antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a colour change that is read at 450 nm in a microplate reader; the colour intensity is proportional to the amount of TIMP1 captured in each well.

The reported assay range is 2.34-150 ng/ml with a sensitivity of <0.92 ng/ml, and the total assay time is 4 h 30 m. The plate is supplied as a pre-coated microplate configured as 12 x 8 well strips and the assay is intended for use with a colorimetric microplate reader. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. An easy-to-follow protocol is provided.

Manufacturer validation data report the following recovery ranges across five samples per matrix: 80-102% (average 91%) in serum, 81-100% (average 90%) in EDTA plasma, and 80-89% (average 84%) in heparin plasma. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 (n=5) was reported as 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.
