Designed for the in vitro quantitative determination of bovine Thrombopoietin in serum, plasma or other biological fluids, this sandwich ELISA from Antibodies.com uses a pre-coated microplate and a biotinylated anti-Thrombopoietin detection antibody followed by an HRP-avidin conjugate, with signal developed using TMB substrate and read at 450 nm in a colorimetric microplate reader.

The assay has a stated range of 78.1-5000 pg/ml with a sensitivity of <31 pg/ml, and a total assay time of 4h 30m. Storage instructions recommend keeping kit components at +4°C or -20°C as indicated, and the product must not be used past its expiration date. Spike-recovery has been evaluated in serum (average 91%, range 80%-102%, n=5), EDTA plasma (average 90%, range 81%-99%, n=5) and heparin plasma (average 84%, range 80%-89%, n=5). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma samples (n=5).

The principle relies on a Thrombopoietin-specific antibody immobilised on the plate to capture Thrombopoietin from standards and test samples. After washing, biotinylated anti-Thrombopoietin antibody binds the captured analyte, and the subsequent HRP-avidin step catalyses the conversion of TMB substrate to a blue product that turns yellow upon addition of acidic stop solution; absorbance at 450 nm is proportional to the Thrombopoietin concentration present in each well and is quantified against the standard curve.
