Bovine Synaptophysin ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of bovine Synaptophysin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit uses the sandwich enzyme immunoassay technique, with an antibody specific for Synaptophysin pre-coated onto a 96-well microtiter plate. Standards and samples are added to the wells and Synaptophysin present in each sample is captured by the immobilized antibody. After incubation and washing, a Biotinylated Anti-Synaptophysin Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualize the reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Synaptophysin captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a working range of 1.56–100 ng/ml and a sensitivity of less than 0.62 ng/ml. It is read on a colorimetric microplate reader with a total assay time of 4 h 30 min. Kit components, including the pre-coated microplate, lyophilized standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, are stored at +4°C or -20°C as instructed. Recovery and linearity performance data are provided for serum, EDTA plasma and heparin plasma samples.
