Bovine Sorbitol Dehydrogenase ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) designed for the in vitro quantitative determination of bovine Sorbitol Dehydrogenase in serum, plasma, tissue homogenates and other biological fluids. The kit, supplied under the Antibodies.com brand, employs the sandwich enzyme immunoassay technique, with an antibody specific for Sorbitol Dehydrogenase pre-coated onto a microplate. Standards and samples are added to the wells and bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-Sorbitol Dehydrogenase Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of an acidic stop solution. The density of yellow is proportional to the amount of Sorbitol Dehydrogenase captured in each well, and the concentration is calculated by reading the absorbance at 450 nm in a microplate reader against a standard curve.

The kit offers a sensitivity of <0.83 U/L and an assay range of 1.56–100 U/L, with a total assay time of approximately 4 hours 30 minutes on a pre-coated microplate read with a colourimetric microplate reader. It is validated for use with serum, EDTA plasma and heparin plasma samples. Reported recovery averages are 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–99%) and 84% for heparin plasma (range 80–89%), each based on n=5. Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is reported as 87–91%, 87–107%, 74–101% and 92–97% respectively for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma. Kit components should be stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date.
