From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of bovine Serum Amyloid A in serum, plasma or other biological fluids. The kit is intended for use with a colorimetric microplate reader, with absorbance read at 450 nm to calculate analyte concentration against a standard curve.

The 96-well microplate is pre-coated with an antibody specific for Serum Amyloid A. Standards and test samples are added to the wells, where Serum Amyloid A is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-Serum Amyloid A Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution visualises the HRP enzymatic reaction, producing a blue product that turns yellow on addition of acidic stop solution; the intensity of yellow is proportional to the amount of Serum Amyloid A captured in each well.

The assay has a sensitivity of <0.054 ng/ml and a product range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Reported recovery in serum (n=5) is 80%–102% with an average of 91%; in EDTA plasma (n=5) is 81%–99% with an average of 90%; and in heparin plasma (n=5) is 80%–89% with an average of 84%. Kit components are stored at +4°C or −20°C as instructed and should not be used past the expiration date.
