Bovine Retinol Binding Protein 4, Plasma ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit is designed for the in vitro quantitative determination of bovine Retinol Binding Protein 4 (RBP4) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate, with detection performed on a colorimetric microplate reader at 450 nm using TMB substrate and an acidic stop solution.

The quantitative sandwich format uses an antibody specific for RBP4 pre-coated onto the wells; standards and samples are added and any RBP4 present is bound by the immobilized antibody. Following washing, a biotinylated anti-RBP4 antibody is added, then HRP-avidin conjugate, and the enzymatic reaction is visualised by catalysis of TMB substrate to produce a blue product that changes to yellow upon addition of the stop solution. The reported product range is 0.625–40 ng/ml, sensitivity is <0.237 ng/ml, and total assay time is 4h 30m. Kit components should be stored at +4°C or −20°C as instructed and not used past the expiration date.

Recovery data from spiked samples (n=5) showed: serum 80–102% (average 91%), EDTA plasma 81–100% (average 90%), and heparin plasma 80–89% (average 84%). Linearity on serial dilution from 1:2 to 1:16 (n=5) ranged 74–107% for serum, 79–108% for EDTA plasma and 82–105% for heparin plasma.
