Bovine Procollagen II N-Terminal Propeptide ELISA Kit

The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for the target pre-coated onto a 96-well microtiter plate.

Standards and test samples are added to the wells, where the analyte is captured by the immobilised antibody. After incubation and washing, a Biotinylated Anti-Procollagen II N-Terminal Propeptide Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction; the resulting blue product turns yellow upon addition of acidic stop solution. The yellow colour density is proportional to the analyte concentration, which is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The kit offers a product range of 78.1-5000 pg/ml, a sensitivity of <34 pg/ml, and a total assay time of 4h 30m. Reported recovery rates are 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma, and 80%-89% (average 84%) for heparin plasma.
