Bovine Platelet Factor 4 ELISA Kit

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit for the in vitro quantitative determination of bovine Platelet Factor 4 (PF4) in serum, platelet-poor plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) and a biotinylated anti-PF4 antibody with an HRP-avidin conjugate; detection is via TMB substrate read at 450 nm on a colorimetric microplate reader, with the yellow product density proportional to PF4 concentration and quantified against a standard curve.

The standard curve range is 0.781–50 ng/ml and sensitivity is <0.27 ng/ml. Total assay time is 4 hours 30 minutes. In spike-and-recovery testing (n=5), recoveries were 80%–102% (average 91%) for serum, 81%–99% (average 90%) for EDTA plasma and 80%–89% (average 84%) for heparin plasma. Dilution linearity (n=5) was demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma.

Kit components are a pre-coated 96-well microplate (-20°C), lyophilized standard (2 vials, -20°C), Detection Solution A (120 μl, -20°C), Detection Solution B (120 μl, -20°C), 30X Wash Buffer (20 ml, +4°C), Sample Dilution Buffer (45 ml, -20°C), TMB Substrate (9 ml, +4°C), Stop Solution (6 ml, +4°C) and 5 adhesive plate sealers. Components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
