A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of bovine uPA in serum, plasma, or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) onto which an antibody specific for uPA has been immobilised. Standards and test samples are added to the wells; following incubation and washing, a Biotinylated Anti-uPA Antibody is added, followed by HRP-Avidin conjugate. A TMB substrate is then used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader.

The reported assay range is 0.156–10 ng/ml with a sensitivity of <0.061 ng/ml and a total assay time of 4h 30m. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The kit is suitable for serum and plasma samples.

Reported recovery values include serum (n=5) ranging 82–102% with an average of 92%, EDTA plasma (n=5) ranging 91–103% with an average of 97%, and heparin plasma (n=5) ranging 86–101% with an average of 93%. Linearity data across dilutions of 1:2, 1:4, 1:8, and 1:16 are provided for serum, EDTA plasma, and heparin plasma samples, with observed ranges of 89–107%, 72–106%, 89–92%, and 88–108% for serum (n=5); 84–102%, 90–99%, 81–110%, and 82–98% for EDTA plasma (n=5); and 90–100%, 80–108%, 91–97%, and 90–107% for heparin plasma (n=5).
