Bovine Parathyroid Hormone Related Protein ELISA Kit

The Antibodies.com Bovine PTHLH ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of bovine parathyroid hormone related protein (PTHLH) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated microplate of 12 x 8 well strips, with a PTHLH-specific antibody immobilised on the wells.

Standards and samples are added to the wells, followed by a Biotinylated Anti-PTHLH Antibody and an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after the addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and PTHLH concentration calculated from the standard curve. The assay has a quantitative range of 15.6-1000 pg/ml, a sensitivity of <6.5 pg/ml, and a total assay time of 4h 30m. Components are stored at +4°C or -20°C as instructed.

Spike-and-recovery testing (n=5) gave average recoveries of 91% in serum (range 80-102%), 90% in EDTA plasma (range 81-100%) and 84% in heparin plasma (range 80-89%). Linearity in serum across 1:2, 1:4, 1:8 and 1:16 dilutions was 87-91%, 87-107%, 74-101% and 92-97% respectively; EDTA plasma gave 90-105%, 84-101%, 90-101% and 79-108%; heparin plasma gave 84-95%, 92-105%, 82-105% and 89-91%.
