Bovine Matrix Metalloproteinase 1 ELISA Kit

A sandwich ELISA (sELISA) from Antibodies.com designed for the in vitro quantitative determination of bovine Matrix Metalloproteinase 1 in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) carrying an immobilised antibody specific for the target; standards and samples are added, followed by a Biotinylated Anti-MMP1 Antibody, an HRP-Avidin conjugate and a TMB substrate. The enzymatic reaction is stopped with an acidic stop solution, producing a yellow colour whose intensity is read at 450 nm in a colorimetric microplate reader and is proportional to MMP1 concentration.

According to the manufacturer, the assay range is 0.156–10 ng/ml and the sensitivity is <0.057 ng/ml, with a total assay time of 4 hours 30 minutes. Components are stored at +4°C or −20°C as instructed; the kit comprises a pre-coated 96-well microplate, a lyophilised standard, Detection Solutions A and B, a 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. The manufacturer also warns not to use the kit past its expiration date.

Reported spike-and-recovery performance gives average recovery of 91% for serum (range 80%–102%, n=5), 90% for EDTA plasma (range 81%–100%, n=5) and 84% for heparin plasma (range 80%–89%, n=5). Dilution linearity is reported across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma.
