The microplate is pre-coated with an antibody specific for LIF, and bound analyte is detected using a Biotinylated Anti-LIF Antibody followed by HRP-Avidin conjugate and a TMB substrate solution, producing a blue product that turns yellow upon addition of the stop solution. Absorbance is read at 450 nm on a colourmetric microplate reader, and the LIF concentration is calculated by reference to a standard curve.

The manufacturer reports a sensitivity of <11.5 pg/ml and an assay range of 31.2-2000 pg/ml, with a total assay time of 4 hours 30 minutes. In spiked recovery testing, average recoveries were 91% for serum (range 80%-102%), 90% for EDTA plasma (range 81%-99%), and 84% for heparin plasma (range 80%-89%). Dilution linearity was assessed at 1:2, 1:4, 1:8, and 1:16 across the same three sample types, with reported recovery values spanning 74%-108% across all dilutions and matrices.
