Description

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of bovine Interferon Gamma Induced Protein 10kDa (IP10) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for IP10 is immobilised on the plate; standards and samples are added and any IP10 present is captured by the coated antibody. After washing, a Biotinylated Anti-IP10 Antibody binds the captured analyte, followed by HRP-Avidin conjugate and TMB substrate. The colour reaction is read at 450 nm on a colorimetric microplate reader and the IP10 concentration is calculated against the standard curve.

The quantitative assay covers a measuring range of 0.156–10 ng/ml with a sensitivity of 0.058 ng/ml, and the total run time is 4 h 30 min. Kit components are stored at +4 °C or –20 °C as instructed and the kit should not be used past its stated expiration date. Reported recovery values are 80–102% (average 91%, n=5) for serum, 81–100% (average 90%, n=5) for EDTA plasma and 80–89% (average 84%, n=5) for heparin plasma.
