A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com for the in vitro quantitative determination of bovine GRP94 (also referred to as Heat Shock Protein 90kDa Beta 1) in serum, plasma, tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, with an antibody specific for GRP94 pre-coated onto a 96-well microplate. Standards and test samples are added to the wells and GRP94 present in each sample is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-GRP94 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The concentration of GRP94 is calculated by reading the O.D. absorbance at 450nm in a microplate reader and referring to the standard curve.

The assay range is 0.781-50 ng/ml and sensitivity is 0.31 ng/ml. Reported sample recovery values average 91% for serum (range 83%-100%), 97% for EDTA plasma (range 91%-103%) and 95% for heparin plasma (range 89%-102%), each measured across n=5. Linearity has been evaluated at dilutions of 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. Total assay time is 4 hours 30 minutes, and detection is colourmetric.

Kit components include a pre-coated microplate arranged as 12 x 8 well strips, a lyophilised standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components should be stored at +4 degrees C or -20 degrees C as instructed, and the kit should not be used past its expiration date.
