This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of bovine heat shock protein 90kDa alpha B1 (HSP90 beta) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids, with reactivity specific to bovine samples. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for HSP90 beta that has been pre-coated onto a 96-well microtiter plate. Standards and samples are added to the captured antibody, followed by a Biotinylated Anti-HSP90 beta Antibody and then an HRP-Avidin conjugate. TMB substrate is used to visualise the reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution; the absorbance is read at 450 nm in a microplate reader and the concentration calculated against a standard curve.

The assay is run on a colourmetric pre-coated microplate platform of 12 x 8 well strips, with an assay time of 4h 30m, a measuring range of 1.56-100 ng/ml and a sensitivity of <0.71 ng/ml. A colorimetric microplate reader is required, and kit components are stored at +4°C or -20°C as instructed, with the kit not to be used past its expiration date.

Sample recovery and dilution linearity have been characterised for serum, EDTA plasma and heparin plasma (n=5 per matrix). Average spike recovery values are 91% for serum (range 80%-102%), 91% for EDTA plasma (81%-101%) and 84% for heparin plasma (80%-89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 has been reported as 70-107%, 89-93%, 86-104% and 80-103% for serum; 95-100%, 85-110%, 81-99% and 90-102% for EDTA plasma; and 77-104%, 88-95%, 94-104% and 80-107% for heparin plasma.
