A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of bovine Haptoglobin in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with a capture antibody specific for Haptoglobin immobilised on the wells. Standards and samples are added, followed by a biotinylated anti-Haptoglobin detection antibody and an HRP-avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm using a colorimetric microplate reader and the Haptoglobin concentration is calculated against the standard curve.

The kit has a stated sensitivity of 41.5 ng/ml and an assay range of 156-10000 ng/ml. The protocol is reported to take 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed. Reported spike-and-recovery averages are 91% for serum (n=5, range 80-102%), 90% for EDTA plasma (n=5, range 81-99%) and 84% for heparin plasma (n=5, range 80-89%), with linearity data provided across 1:2 to 1:16 dilutions for serum, EDTA plasma and heparin plasma.
