Bovine Galectin 8 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of bovine Galectin 8 (Gal-8) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated microplate (12 x 8 well strips) with an antibody specific for Galectin 8/Gal-8 immobilised on the wells. Standards and samples are added, the plate is washed, and Biotinylated Anti-Galectin 8/Gal-8 Antibody is then added, followed by HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution. The density of yellow is proportional to the amount of Galectin 8/Gal-8 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

The quantitative measurement range is 0.312-20 ng/ml with a sensitivity of <0.074 ng/ml. Total assay time is 4h 30m, and kit components are stored at +4°C or -20°C as instructed. Spike-and-recovery performance was demonstrated with serum (n=5) ranging 80-102% (average 91%), EDTA plasma (n=5) ranging 81-100% (average 90%), and heparin plasma (n=5) ranging 80-89% (average 84%). Dilution linearity across 1:2 to 1:16 dilutions was also reported in the same sample types.
