Bovine Fibroblast Growth Factor 4 ELISA Kit

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (ELISA) for the in vitro quantitative determination of bovine Fibroblast Growth Factor 4 (FGF4). The kit is intended for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, and employs a pre-coated microplate (12 x 8 well strips) read on a colorimetric microplate reader at 450 nm.

In the workflow, standards and samples are captured by an immobilised anti-FGF4 antibody, then detected sequentially with a Biotinylated Anti-FGF4 Antibody and an HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow on addition of the acidic stop solution; the yellow intensity is proportional to FGF4 concentration and is quantified against the standard curve. The total assay time is 4h 30m, and kit components are stored at +4°C or -20°C as instructed.

The standard curve spans a product range of 31.2–2000 pg/ml with a stated sensitivity of <13.8 pg/ml. Reported recovery in serum (n=5) is 80%–102% (average 91%), in EDTA plasma (n=5) is 81%–100% (average 90%) and in heparin plasma (n=5) is 80%–89% (average 84%). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16, with serum recoveries of 87–91%, 87–107%, 74–101% and 92–97% respectively.
