From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of bovine Cathepsin D in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The 96-well microtiter plate is pre-coated with Cathepsin D antigen and uses a competitive enzyme immunoassay technique in which Cathepsin D present in samples or standards competes with the immobilized antigen for binding sites on a Biotinylated Anti-Cathepsin D Antibody. Following incubation and washing, an HRP-Avidin conjugate is added, and TMB substrate solution is used to visualize the reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is inversely proportional to the amount of Cathepsin D present, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against the standard curve.

The reported product range is 3.12–200 ng/ml with a sensitivity of <0.88 ng/ml, run on a pre-coated microplate platform of 12 x 8 well strips with a total assay time of 2h 30m, read on a colorimetric microplate reader. According to the manufacturer's validation data, average recovery in undiluted samples ranges from 84% (heparin plasma) to 91% (serum), and recovery remains broadly consistent across 1:2 to 1:16 dilutions. Kit components should be stored at +4°C or -20°C as instructed. The product is sold for research use only.
