Designed by Antibodies.com for the in vitro quantitative determination of bovine Aprotinin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, this competitive ELISA kit employs the competitive enzyme immunoassay technique. A 96-well microtiter plate is pre-coated with Aprotinin antigen. During incubation, Aprotinin present in samples or standards competes with immobilized antigen for binding sites on a biotinylated anti-Aprotinin antibody; the more analyte present, the less antibody binds to the plate. After washing, an HRP-avidin conjugate is added and TMB substrate is used to visualize the reaction, producing a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and is inversely proportional to Aprotinin concentration.

The assay covers a standard range of 1.56–100 ng/ml with a sensitivity of <0.43 ng/ml, runs in approximately 2h 30m, and is performed on a pre-coated microplate platform configured as 12 x 8 well strips. Reported average recovery values are 92% (range 82–102%) from serum, 95% (90–100%) from EDTA plasma, and 94% (88–100%) from heparin plasma.

Components include a pre-coated 96-well microplate, lyophilized standard, Detection Solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers; components are stored at +4°C or -20°C as instructed.
