Bovine Alpha-2-Plasmin Inhibitor ELISA Kit

A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of bovine alpha 2 Antiplasmin in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for alpha 2 Antiplasmin pre-coated onto a microplate (12 x 8 well strips). Standards and test samples are added to the wells, followed by a Biotinylated Anti-alpha 2 Antiplasmin Antibody and an HRP-Avidin conjugate. Detection uses a TMB substrate solution visualising the HRP enzymatic reaction, which changes colour upon addition of an acidic stop solution, with absorbance read at 450nm on a colorimetric microplate reader. The concentration of alpha 2 Antiplasmin is calculated by reference to a standard curve.

The assay has a reported sensitivity of 7.7 ng/ml and an assay range of 15.6-1000 ng/ml, with a total assay time of 4h 30m. Sample recovery was evaluated as follows: serum (n=5) ranged from 83% to 100% with an average of 91%; EDTA plasma (n=5) ranged from 90% to 103% with an average of 96%; and heparin plasma (n=5) ranged from 90% to 99% with an average of 94%. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma samples.

Kit components include a Pre-Coated 96 Well Microplate, Lyophilized Standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and Plate Sealers. Components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
