The assay uses an anti-Akt1 (phospho Tyr474) capture antibody followed by HRP-conjugated secondary detection, producing a colorimetric signal on a tissue culture treated microplate, with a total assay time of 4 hours 30 minutes.

Three normalization approaches are described in the protocol: an included Anti-GAPDH Antibody serves as an internal positive control, Crystal Violet whole-cell staining is used to determine cell density after the colorimetric readout, and an Anti-Akt1 Antibody is provided so that absorbance values for the non-phosphorylated target can be used to normalize those of the phosphorylated target.
